In 1983, in a US Department of Energy laboratory in the town of Hawkins, Indiana, a scientist is attacked by an unseen creature. 12-year-old Will Byers vanishes after encountering the creature while riding his bicycle home from a Dungeons & Dragons session with his friends. The next day, a young girl with a shaved head, wearing a hospital gown, steals food from a local diner. The owner, Benny (Chris Sullivan), takes pity on her and feeds her before calling social services. A woman posing as a social worker arrives and shoots Benny. Armed men search the diner for the girl, but she escapes. Joyce, Will's mother, believes she hears Will's voice on a distorted phone call, but her phone short circuits. Will's friends Lucas, Mike, and Dustin find the girl in the woods as they search for Will.

While working on a commercial, Dev finds out that a colleague had been pursued by a man on her way home. When Dev tells Denise and Rachel about it, the women share similar experiences. Dev realizes he is privileged just by being male, and he identifies himself as a feminist. While hanging with the cast of his commercial one night, he discusses with the director that all the women in the commercial are just there for eye candy. The director revises the commercial so that all the main speaking roles are given to the women, and Dev is fired for not fitting into his role. Although he is disappointed, the women are thankful that he stood up for them.

Fiona has a tryst with the Axman, which is complicated as the side effects of her chemotherapy begin to show. Zoe reattaches Spalding's tongue and interrogates him, killing him after he admits Fiona was Madison's real murderer. Madison and Kyle bond over their lack of feeling as undead, and they have sex, which Zoe inadvertently witnesses. Madison, sympathetic to Zoe's feelings, proposes they share, and they have a threesome with Kyle. Delphine and Queenie bond over hamburgers, but Delphine tells Queenie that the other witches will never truly accept her because of her race. Queenie visits Marie, who offers to let Queenie join the voodoo practitioners if she brings her Delphine. Delphine confesses to Queenie that she murdered a child her husband fathered with a slave and used its blood as a nightmask; disgusted, Queenie turns Delphine over to Marie.

Jim meets Hallie Shea (Grace Gummer), an embedded reporter with the Romney campaign. Charlie Skinner takes Will off the 9/11 tenth anniversary story because of his American Taliban comment, and Will is condemned on the House floor as a result. Maggie and Sloan track down the person who posted the youtube video of Maggie's confession about Jim in front of the Sex and the City bus and make a deal with her to take it down. However, Maggie's best friend Lisa, (Kelen Coleman) soon discovers the video and decides to end their friendship. Maggie wants to do a story in Africa and pitches it to Mac. Don tries to convince Will to strengthen coverage of the Troy Davis execution. Jerry Dantana tells Mac about Operation Genoa, a mission conducted by Marine Special Operations (MARSOC) to rescue two captured Marines by using sarin gas against the enemy. Neal is arrested while filming an Occupy Wall Street protest leading to Will going to the police station to bail him out. Takes place between August 25 and September 21, 2011.

Trashy neighbor Rita Glossner (Brooke Shields) returns and is about to rain fire upon the Heck household when she accuses Frankie of stealing her hose. Meanwhile, Sue takes a peek at Mike's paycheck and is so shocked at how little he makes that she decides to call off her first official road trip to Cincinnati for her school mascot duties in order to save money; and while Brick learns about the birds and bees in school, Axl shocks his little brother by explaining the whole process in graphic detail.

Classes resume at Greendale Community College and the group enroll in a new class, Anthropology 101, which is taught by June Bauer, an esteemed, but quirky anthropology professor. Jeff deals with the aftermath of last season's dance after Britta confessed her love for him, and he later shared a secret kiss with Annie. Senor Chang decides to enroll at Greendale as a student.

J.D. gets called in to work on his day off, but the only thing he wants to do is get home. Meanwhile Elliot tries to convince everyone she is an expert on endocrinology following her brief fellowship, Carla has doubts over being a parent, and Turk has to wrestle over an issue that will either help his career or break his conscience.

Stan finds Deputy Director Bullock, his supervisor, starting a relationship with Hayley. However, he can't risk saying or doing anything about it because he's up for a promotion at the CIA. Meanwhile, Steve finds Dick Cheney's BlackBerry and uses the contact numbers to play pranks on political figures.

Molly returns to Sacred Heart, and J.D. blows off his girlfriend Kylie to go out with her. Carla tries not to make "mountains out of molehills" when she realizes Turk is still in contact with an ex-girlfriend. The Janitor bets Dr. Cox that he can get a date with Elliot, the wager being his van against Dr. Cox's Porsche.
