June moves to New York City, but when the company she was going to work at goes under, she gets a job at a coffee shop with her would-be boss Mark. She moves into Apartment 23 with the cunning Chloe, who is friends with ex-boyfriend and actor James Van Der Beek, and lives across the street from the perverted Eli who masturbates to June. Their neighbor Robin warns June to not "trust the bitch in Apartment 23," which turns out to be good advice, as Chloe tries to scam June out of her money. June retaliates by selling all of Chloe's stuff, but then has to get her ottoman back (because it has energy pills sewed into it). Chloe then proves to June that her fiancee was cheating on her by getting him to almost hook up with her. June is sad, but she and June bond over it, and discover the beginning of an unusual friendship.

Kurt fixes up his father on a date with Finn's mother, Carole, in hopes of having a regular family and of getting closer to Finn. His plan backfires when his father bonds with Finn over sports, leaving Kurt feeling excluded. Will rents out April Rhodes's roller rink for New Directions as a temporary practice space. Will helps April sort out her life once more, culminating in April becoming a millionaire and buying the school auditorium for the glee club. Mercedes deals with weight issues after Sue tells her to lose ten pounds for an upcoming Cheerios interview, but learns to feel comfortable with who she is.

When famed neurosurgeon Dr. Ginsberg and her team pay a visit to St. Ambrose to wake Kayla from her coma, Addison is shocked to see Amelia Shepherd - Derek's younger sister - on the team. After Dr. Ginsberg declares nothing can be done, Amelia causes strife among Addison, Sam and Pete when she claims she can save Kayla's life. Charlotte seeks Cooper's help with a sex education seminar for the elderly that only leads to more tension between the two, as they try to figure out how to work together.

Will, trying to find a twelfth member in the wake of Rachel's defection to the school musical, recruits former glee club star April Rhodes. Finn flirts with Rachel continuously in an attempt to convince her to return, hoping that having her in the club will increase his chances of gaining a music scholarship. While Finn takes Rachel bowling, they kiss briefly after Rachel bowls a strike. Although Rachel is angry when she discovers that Quinn is pregnant, she ultimately rejoins the club. Will confesses to once having had a crush on April, and encourages her to sober up and pursue her dreams of performing on Broadway. The glee club performs at Invitationals, launching themselves onto the show choir competition circuit.

When Izzie's daughter is brought to the hospital for leukemia treatment, Izzie is asked to donate her bone marrow in support. Derek finds that Meredith stands in his way of becoming chief. Marlowe continues to intrude on Burke and Cristina's lives with an impending surgery Burke will perform. Meredith becomes closer to her stepmother. Parents claiming to be Ava's come in to identify her but learn she is not theirs.
